บทคัดย่อ (EN): Thalassemia is the most common genetic disorder in Thailand. Beta-thalassemia is caused by reduced or absent of ß-globin resulting in insufficient adult hemoglobin (HbA, a2ß2) production. One of the most effective strategies to reduce severe anemia in patients is an increasing level of fetal hemoglobin (HbF, a2g2) to compensate the lack of HbA. This study, we aimed to develop a cell-based assay system for searching the active substances inducing g-globin and HbF expression. The cell model was designed and established for the detection of HBG (g-globin gene) promoter-activating activity associated with g-globin expression. This system was used to test over 180 extracts from medicinal plants. Of these, 7 plants extracts exhibited the high HBG promoter-activating activity. Five active substances, belong to flavonoid and benzofuranone classes, were able to isolate from these plant extracts. The substances were investigated for g-globin mRNA and HbF levels by quantitative PCR and HPLC-PDA analysis, respectively. The active substance T01 elevated g-globin expression and increased HbF level to 1.7-2.8 folds in K564 cell line. Moreover, F-cell induction was revealed in beta-thalassemia derived erythroid progenitor cells treated with T01, suggesting that T01 would be a promising HbF inducer to use as medical aids for beta-thalassemia patients.
