บทคัดย่อ: The 4 samples of button mushroom fruit bodies from various places in Nakhon Pathom province, and Bangkok, were collected, and the pure mycelia were isolated by culturing those mushroom tissues on PDA media. Seven selected isolates of button mushroom were BQ2/1, CQ3/2, CBL46, NBK46, NBK46.1, AL2 and AS13. The genetic diversity of those button mushroom strains was performed by comparing the DNA sequences in the ITS region of rDNA, and the genetic relationships of each strain using PHYLIP program. The results indicated that the DNA sequences in the ITS region of all 7 isolates of button mushroom were similar to those sequences of Agaricus spp., A. bitorquis และ A. pseudopratensis , the similarity of DNA in the ITS region of those mushrooms was 77-99%. From molecular phylogenetic analysis, those button mushrooms could be divided into 3 groups. The first group consisted of isolate AL2 and NBK46.1, the second one was isolate AS13, BQ2/1 and CQ3/2, and the last group was isolate CBL46,NBK46. ? The study of suitable carbon sources, nitrogen sources and pH on mycelia growth of button mushroom isolate CQ3/2 was performed. The results indicated that the most suitable carbon sources for mushroom mycelia growth were glucose and maltose. The nitrogen sources were yeast extract, peptone and malt extract, and the optimum pH was in the range of 6.15 (control) to 8.0. The cultivation of button mushroom isolate CQ3/2 was performed in polypropylene bag, using substrate 3 formulae, which were spent para-rubber sawdust from Hugarian oyster mushroom production: new para-rubber sawdust in the ratio of 1:1, 1:2 and 1:4 by dry weight. The results showed that the mushroom mycelia provided full grown in the first formula substrate (ratio 1:1) only in small bag (600 grams/bag), and the mycelia could grow in all 3 formulae in big bag (1,200 grams/bag) within 120 days. The average yields were 243.75 grams per small bag, in 94 days, after casing with peat moss plus 6.0% lime, pH 7.99. The substrate having each formula had no influence on those yields in big bag, which were in the range of 35.47-89.26 grams per bag within 24 days. The study of spawn quantities, and the cultivation of button mushroom in various containers, such as plastic bag (1,200 grams substrate), plastic basket size 25.5x30.5x7.5 cm (1,800 grams), big and small plastic bucket, which had diameter and height of 15 and 11.5 cm (600 grams), and 11 and 10 cm (400 grams) was conducted. The substrate used was formula 1, which modified by adding dry horse manure and fertilizer ammonium sulfate, or the fertilizer 16-20-0. The results indicated that the amount of 10, 20 and 30 grams spawn per bag (1,200 grams) had no effect on the time for full grown mycelia, which was 88 days. But the application of 10 and 20 spawn gave average high yield of 156.33 and 110.55 grams per bag within 44 days. Moreover, the cultivation of button mushroom in basket and big bucket gave the average yield of 112.15 grams per basket, and 111.20 grams per bucket. Whereas, the cultivation in plastic bag and small bucket gave the average yield only 50.35 and 38.20 grams within 47 days.
