บทคัดย่อ (EN): Sampling of Hevea leaves exhibiting symptoms of Corynespora leaf fall disease was carried out in rubber plantations in Malaaysia. Twenty one (21) isolates of the fungus C. cassiicola obtained from the 1998 -2005 collections were used in this study. The isolates were purified using single spore isolation method and cultured on potato sucrose agar (PSA) for diversity analysis. Differences in colony size, growth, colour (slight to dark grey on top; brown to black at bottom), shape (round to slightly polygonal) and texture (thin to thick hairy) were observed among the isolates. The isolate CKTO5D showed the fastest growth rate and the biggest colony size at seven days after cultured (70.8 mm in diameter). This isolates stands separately to other in means separation analysis using Duncan multiple range test. The conidia morphology and dimensions were recorded from detached leaf inoculations by using Pixelink Megapixel Firewir Camera model PL- A662 lixed on Olympus Microscope BH-2 and measured usingVideoTest Size 5 software (100 conidia /isolated). Marked differences in conidial dimensions, such as in shapes (obclavate, cylindrical or oval; straight or curved), sizes (17 um- 942 um long; 3.9 um -16.8 um wide) and the number of pseudoseptate (0-70), were observed among the isolates. Occurrence of variability in conidial shapes and sizes within the sameisolate were also not uncommon. However, the peculiar characteristics of each isolate enabled differentiation of the analysed isolates. Isolate CKTI contained the longest average conidial length (395.5 um) and the highest average number of pseudoseptate(24.2), whereas the largest average conidial width (13.0 um) was observed in isolated CLKKI. Based on the means separation analysis, the isolates were separated into 10 groups for spore length: 11 groups for number of pseudoseptate, with CKTI as the stand alone group; and 9 groups for spore width, with CLKKI as the stand alone group. Classification of the C. cassiicola isolates will be complemented with molecular marker analyses.
