บทคัดย่อ (EN): Developing a recycling process of Natural Rubber (NR) is of great interest and expected to reduce the wastes of NR products and biodegradation process to reuse NR polymers as alternative biomass serves as a key technology without a burden on environment. Bacterial degradation of NR has been well characterized by gram-negative bacteria, Xanthomonas sp. Strain 35Y the has RoxA for rubber oxygenase, which is responsible to the cleavage of NR. A screening of unique bacteria for NR degradation has been performed and several rubber-degrading bacteria were isolated in Japan. A NR-degrading gram-negative bacterium, Rhodobacter gummiphilus strain NS21 degrades deproteinized NR (DPNR) into aldehydes. NS21 has two enzyme genes, latA1 and latA2, that have weak amino-acid-sequence similarity of 35% and 65% respectively, to RoxA. Similarity between LatA1 and LatA2 is 37% in amino acid-sequence. The latA1 mutant strain lost the ability to convert DPNR into aldehydes but is retained clearing-zone-forming ability that is thought to be derived from the structural conversion of DPNR. The latA2 mutant and latA1A2 double mutant strains lost both ability, suggesting the latA2 gene is involved in the clearing-zone-formation during DPNR degradation. To demonstrate the functional interaction of LatA1 and LatA2 for the DPNR degradation process, cultivation of each mutant with wild type or each mutant was performed. The conversion of DPNR into aldehydes by the latA2 mutant was detected only when it was cultivated adjacent to wild type or latA1 mutant that had innate latA2 genes. These results suggest that both the latA1 and the latA2 genes are indispensable for DPNR degradation by NS21.
