บทคัดย่อ (EN): The berberine bridge enzyme (BBE) (EC1.5.3.9), an enzyme associated with plant defence and the stress response. To examine the protective roles of this enzyme in the rubber tree (Hevea brasiliensis) that is commonly subjected to multiple physiological stresses, the HbBBE1 gene was cloned from the leaf of the rubber tree. Changes in gene expression under different experimental treatments were examined. The results showed that the full length cDNA of HbBBE1 is 1757 bp in length; it contains an open reading frame (ORF) of 1620 bp which encodes 539 amino acids, a 135 bp 3’untranslated region and a 3 bp 5’untranslated region. The molecular mass of the putative HbBBE1 protein is estimated at 60.2 kDa with a theoretical pI of 7.53. The predicted protein of HbBBE1 contains a signal peptide, a transmembrane region, an FAD binding domain, and a BBE domain which is found specifically in the berberine bridge enzymes and berberine bridge-like enzymes. HbBBE1 shared the highest similarity with the BBE protein from Manihot esculenta, with 80.1% identical residues. The HbBBE1 gene was found to be preferentially expressed in the latex, but gene expression was also observed in bark, leaves and flowers. Expression of the gene was upregulated by mechanical wounding both in leaves and in latex collected from 7 years old virgin (previously untapped) trees. On the other hand, gene expression was down regulated in the latex from virgin trees by methyl jasmonate (MeJA). Gene expression was also increased in leaves infected by powdery mildew fungus (Oidium heveae Steimnann). As HbBBE1 expression was rapidly induced by wounding, drought, light, and hormones treatments, the gene is believed to be involved in in multiple defense responses, including a reaction to leaf fungal infection.
