บทคัดย่อ: การศึกษานี้มีวัตถุประสงค์ในการศึกษาประสิทธิภาพ Fluorocult LMX Broth Modified ในการตรวจวิเคราะห์ Total Coliforms และ Escherichia coli ในตัวอย่าง 3 กลุ่ม คือ น้ำและน้ำแข็ง กลุ่มสัตว์น้ำ และกลุ่มผลิตภัณฑ์สัตว์น้ำ โดยเปรียบเทียบกับวิธีมาตรฐาน BAM Online (2020) ผลการศึกษา Relative Trueness โดยการทำ Scatter Plot พบว่าผลการวิเคราะห์อยู่บนเส้นหรืออยู่ใกล้เส้น Line of Identity ส่วนการทำ Bland-Altman Difference Plot พบว่า ค่าเฉลี่ยของผลเปรียบเทียบระหว่างสองวิธีอยู่ในขอบเขตของเส้นขีดจำกัดบนและขีดจำกัดล่าง ที่ระดับความเชื่อมั่น 95 เปอร์เซ็นต์ ตามเกณฑ์กำหนด แสดงให้เห็นว่าการใช้ Fluorocult LMX Broth Modified ให้ผลสอดคล้องกับวิธีมาตรฐาน ส่วนผลการศึกษา Accuracy Profile โดยใช้ตัวอย่างน้ำดื่ม และกุ้งต้มแช่แข็ง พบว่าผลทดสอบ Total Coliforms และ Escherichia coli อยู่ในช่วงเกณฑ์การยอมรับ +0.5 Log แสดงว่าการใช้ Fluorocult LMX Broth Modified ให้ผลการทดสอบถูกต้องสามารถยอมรับได้และมีความเท่าเทียมกับวิธีมาตรฐาน การทดสอบความจำเพาะเจาะจงของการใช้ Fluorocult LMX Broth Modified ต่อการตรวจพบเชื้อจุลินทรีย์เป้าหมายและไม่ใช่เป้าหมาย พบว่ามีความสามารถตรวจพบเชื้อเป้าหมายได้ 100 เปอร์เซ็นต์ และไม่สามารถตรวจพบเชื้อที่ไม่ใช่เป้าหมาย 100 เปอร์เซ็นต์ เช่นเดียวกับวิธีมาตรฐาน ส่วนผลศึกษา Estimated Bias (eBias) Determination มีค่าไม่เกิน +0.5 Log จึงสามารถนำ Fluorocult LMX Broth Modified มาใช้ในการตรวจวิเคราะห์ในห้องปฏิบัติการได้โดยให้ผลการทดสอบที่ถูกต้องและมีความน่าเชื่อถือ นอกจากนี้ผลการเข้าร่วมทดสอบความชำนาญกับหน่วยงานภายนอก ในปี พ.ศ. 2566 และปี พ.ศ. 2567 ในตัวอย่างน้ำและ Lyophilized Cultures พบว่า มีค่า Z- score น้อยกว่า 2 แสดงว่าผลการทดสอบยอมรับได้ สำหรับค่าใช้จ่าย และระยะเวลาการตรวจวิเคราะห์ กรณีตรวจไม่พบเชื้อตั้งแต่ขั้นตอน Enrichment วิธีมาตรฐานจะใช้งบประมาณในการวิเคราะห์น้อยกว่า ใช้ระยะเวลาในการทดสอบ 1-2 วัน กรณีตรวจพบเชื้อและต้องยืนยันผลการทดสอบวิธีมาตรฐานจะใช้งบประมาณ ระยะเวลาและแรงงานในการทดสอบมากกว่าการใช้ Fluorocult LMX Broth Modified
บทคัดย่อ (EN): This study aimed to study the efficiency of Fluorocult LMX Broth Modified for Total Coliforms and Escherichia coli in 3 groups of samples: water and ice, fisheries and fisheries product by comparing with the standard method BAM Online (2020). The results of the Relative trueness study by doing a Scatter plot found that the analysis results were on the line or near the line of identity. As for doing the Bland-Altman Difference plot, the average comparison results between the two methods fell within the upper and lower limit lines at 95% confidence level, indicated that Fluorocult LMX Broth Modified provided results consistent with the standard method. The Accuracy Profile study was conducted by drinking water and frozen boiled shrimp samples, found that the test results fell within the acceptable range of +0.5 Log, indicated that Fluorocult LMX Broth Modified could be accepted as having accuracy comparable to the standard method. It also demonstrated 100% inclusivity and exclusivity in testing. The Estimated Bias (eBias) Determination study showed a value not exceeding +0.5 Log, confirming that Fluorocult LMX Broth Modified can be used in laboratory analysis with accurate and reliable test results. Additionally, the proficiency testing results with external agencies in 2023 and 2024 in water samples and lyophilized cultures found that Z-score value were less than 2, indicated that the test results were acceptable. Regarding costs and analysis time, if no organisms were detected during the enrichment step, the standard method was less expensive and required 1-2 days for testing. If organisms were detected and confirmation was required, the standard method required more budget, time, and labor to test than Fluorocult LMX Broth Modified.
