บทคัดย่อ (EN): Characterization of bacterial rubber degrading enzyme genes will lead to the use of recombinant enzymes and application of rubber degradation products for new carboxylated cross- linking polymers and new processing system for surface fabrication. We are focusing on a latex clearing protein (lcp) from the Nocardia sp. NVL3 which was isolated from wastes of a rubber processing factory in Thanh Hoa province, Vietnam. Thus, an lcp in strain NVL3 was cloned in a cloning vector and was heterologously expressed in E. coli BL21 (DE3) resulting a recombinant BL21::pEHIcpH. In this study, we are successful to purify His-tagged Lcp from soluble fraction of BL21::pEHIcpH cells and separated it in a SDS - Polyacrylamide gel. In order to analyse peptide mass fingerprint, in-gel 47.3 kDa protein then was purified and digested with Trypsin. As the results of Matrix-assisted laser desorption/ionization - Time of flight mass spectrometer (MALDI-TOF/MS), all peptides of Lcp were separated in sequence and computed for mass. To confirm the enzymatic activity, we carried out the reaction in vitro by using 0.4 mg of purified His-tagged Lcp with in-flask coated synthetic poly (cis-1,4-isoprene) and flask samples were incubated for 24 hours at 30◦C or at 37◦C.Gel permeation choromatography (GPC) analysis of those samples show the degeneration of a peak derived from poly (cis-1,4-isoprene) and the arising of the another peak at higher retension time. By that, we strongly comment Lcp itself cleaves rubber polymer to lower molecular weight products. In contrast, no rubber degradation lead by extracellular proteins of BL21::pEHLcpH culture can be observed. Consistently, Schiffs staining in a paper disk assay revealed that purified Lcp and soluble protein fraction from BL21::pEHLcpH cells can accumulated Aldehydes persistently on the rubber plate. Thus, our study indicated Lcp is involved in rubber degradation of strain NVL3 by cleavage of rubber polymer into lower-molecular-weight aldehyde-containing compounds. This protein can be purified highly for detail enzymatic characterization and further applications on rubber processing can be acquired.
