บทคัดย่อ (EN): Colletotrichum leaf disease of rubber tree caused by C. gloeosporioides is one of the major causes of declining yields of rubber tree. Little is known about the fungal molecular characters that are important for pathogenicity and fungicidal resistance of it on rubber tree. In this study, we cloned the CgPbs2 gene, the key component of the Hog1 pathway which controls various aspects of osmoregulation and fungicidal resistantce in various fungal pathogens, in the causal agent of Colletotrichum leaf fall disease of Hevea brasiliensis. We characterized the function of CgPBS2 gene by loss of function method. Based on the Hog1 pathway plays a important role in stress responses, we obtained the CgPbs2 gene deletion mutants by PEG mediated transformation of protoplast after reduced the concentration of sucrose in screening medium from 1.0M to 0.2M. Then the complementary transformants and GFP-labbed CgPbs2 gene transformants were selected directly under the high hyperosmotic medium( PDA+1.5M Sorbitol) without using other selection gene marker. The phenotype observation results showed that CgPBS2 protein mainly localized in cytoplasm in conidia of the CgPbs2-GFP transformants. And disruption of CgPbs2 leads to sensitivity to hyperosmosis and high salt, and resistance to the fungicide fludioxonil. About virulence, there was no obvious difference were observed between null mutant and wild-type strain. These results provide the first insights about that elucidate the role of the CgPbs2 gene in osmotic and salt stress and fungicide fludioxonil resistance using the osmotic stress sensibility as selection marker.
