บทคัดย่อ (EN): Corynespora cassiicola is an important plant pathogen causing Corynespora leaf fall (CLF) disease of the rubber tree. Diversity analysis based on the virulence level and Ribosomal DNA Internal Transcribed Spacer (rDNA-ITS) region sequence were undertaken to investigate the effect of geographic and host rubber clones of isolates to virulence level of 23 C. cassiicola isolates from six rubber clones (RRIC 100, BPM 24, BPM 1, PB 260, GT 1 and RRIM 600) and as well as to observed the evolution pattern of the isolates. Virulence analysis showed that there was difference of virulence level of each isolates to the six test clones. Geographic and host rubber clones origin of isolates influenced the virulence level and rDNA-ITS sequence variation of the isolates analyzed. Compared to geographic origin, host rubber clones had more clear influence on virulence level of isolates. The diversity analysis of rDNA-ITS sequences showed that there were three points of Single Nucleotide Polymorphism (SNP) on the 23 isolates analyzed. It separated the isolates into 5 of different haplotypes. Most of the isolates belonged to the haplotype 1. There was an inter-relation between rDNA-ITS sequences with the virulence, Changes in the base of SNPs caused change in the virulence level of isolates. Phylogenetic analysis of the 23 isolates and rDNA-ITS sequence from database demonstrated that C. cassiicola isolates have a high genetic diversity. However the clustering did not indicate the country and host species origin of isolates. The information obtained in this study will be very useful in developing strategies for disease management and breeding for resistance to CLF disease.
