บทคัดย่อ (EN): Cases of mushroom poisoning in local Thai populations have increased annually and continue to be a problem faced by health care professionals. A total of 5,082 cases with 27 deaths were reported to the Bureau of Epidemiology, Ministry of Public Health during 2014 to April 2018. In the laboratory, poisonous mushrooms are routinely identified based on morphology and toxic chemistry. However, the samples are generally not well preserved in for clinical diagnosis and standard methods for morphological identification do not always yield satisfactory results as well as standard reference material is expensive. Hence, the aims of this study were i) to construct a DNA database of nuclear ribosomal internal transcribed spacer (ITS) region, nuclear large-subunit (LSU) ribosomal DNA, DNA-directed RNA polymerase II subunit rpb1 (RPB1), Amatoxins-encoding gene (AMA) and Phallotoxins-encoding gene (PHA) and ii) to investigate the applicability of DNA markers as a DNA barcode for identifying the poisonous mushrooms and transfering knowledge-based prevention to communities. A total of 312 new DNA sequences of mushroom samples obtained from clinically reported cases and natural habitat were generated in this study. BLAST database with local sequences were constructed in Toxicology center as a references. Molecular barcoding confirmed the presence of new species and new records of several poisonous mushroom species, namely Amanita digitosa (Gastrointestinal syndrome), A. exitialis (Phalloides syndrome), A. gleocystidiosa (Phalloides syndrome), Cantharocybe virosa (Gastrointestinal syndrome)and Chlorophyllum globosum (Gastrointestinal syndrome). Four unidentified species belong to the genera Hygrocybe, Inocybe, Parvixerocomus and Tylopilus, which are responsible for gastrointestinal syndrome, muscarine syndrome and the later undefined species of Tylopilus and Parvixerocomus are responsible for a cause of unexpected death.These poisonous mushroom species provided new and informative data for clinical studies. Measures should be taken to prevent the consumption of toxic mushrooms.
