บทคัดย่อ (EN): Cellulose is naturally-occurring plant cell wall constituents and is the most and second most abundant polysaccharides on earth. An economically viable process for converting plant biomass to fermentable sugar is a prerequisite for the development of cellulosic biomass-based bio-refineries for the production of food, feed, chemicals, and fuels. Cellulose can be converted to fermentable sugar by microbial cellulose degrading enzymes, called as cellulose. Filamentous fungi, including Trichoderma, Aspergillus, Penicillium and Acremonium produce significant amounts of cellulose. The enzymatic hydrolysis of cellulose usually involves endo-β-1,4-glucanases (EG, EC 3.2.1.1), which randomly cleave internal bonds of amorphous cellulose, cellbiohydrolases (CBH, EC 3.2.1.1.91), which release cellobiose as a major product from the ends of cellulose chains, and beta-glucosidases (BGL, EC 3.2.1.21), which convert cellobiose and cello-oligosaccharide into glucose. Rubber-wood is one of the main industrial crops in South East Asia. After yielding of their latex the waste rubber trees could be utilized as the renewable carbon resource by using microbial cellulolytic enzymes. However, plant cell wall has complex structure composed of polysaccharides such as cellulose, hemicelluloses, and pectin, and a complex organic polymer, lignin. Therefore, for effective enzyme degradation, physical, chemical, or biological pretreatment is necessary. In this work, several pretreatment methods were tested to achieve effective enzymatic degradation. The waste rubber tree was milled to obtain fine powder. The fine powder of waste rubber tree was treated by sulfuric acid, sodium hydroxide, or hydrothermal treatment. Each treated sample was subjected to enzymatic degradation by using cellulose preparations of cellulolytic filamentous fungus Trichoderma reesei. Consequently, it was revealed that sulfuric acid pretreatment was effective for cellulolytic reaction and final conversion ratio of cellulose into glucose was over 55%.
