บทคัดย่อ (EN): Understanding of heat stress responses can be useful for elucidate heat tolerance mechanism. This study examined the effect of high-temperature stress on physiological responses and expression levels of Hsp72 (inducible form) and Hsp73 (constitutive form) in cattle with different heat-tolerant capacities, including Bos indicus (Sahiwal; n=5), Bos taurus (Holstein Friesian (100%HF); n=5), and Bos taurus/Bos indicus crosses (50% Holstein Friesian (50%HF); n=5 and 87.5-97.7% Holstein Friesian (>87.5%HF); n=5). The physiological responses recorded include rectal temperature, respiration rate, and sweating rate, at 0600 (THI=72.12), 0900 (THI=80.32), 1200 (THI=90.40), and 1500 (THI=94.00) in summer (March 2011). Leukocytes were collected to determine the level of Hsp72, and Hsp73 by western blot analysis. The results indicated that THI were highly affected in rectal temperature, respiration rate, and sweating rate (P<0.01). Changes in rectal temperature in Sahiwal and 50%HF were less than >87.5%HF and 100%HF while changes of respiration rate in 50%HF, >87.5%HF and 100%HF were higher than in Sahiwal. The sweating rate of Sahiwal was found to be significantly higher than 50%HF, >87.5%HF and 100%HF. The level of Hsp72 of 50%HF, >87.5%HF, and 100%HF increased in response to THI, especially when THI was higher than 90, the Hsp72 level in 100%HF decreased. In Sahiwal, the level of Hsp72 was slightly increased at the lower THI but increased sharply when THI was higher than 90. The expression level of constitutive Hsp73 protein in leukocytes of all cattle was not affected by THI but Sahiwal showed higher level of Hsp73 than other group of cattle. These results illustrated that there were differences in heat stress responses in both the physiological indicators and level of Hsp72 of the different heat-tolerant capacity cattle.
