บทคัดย่อ: The Aedes aegypti mosquito is the primary vector of dengue fever and zika fever. In Thailand, the diseases have become major public health problems. Although there is currently dengue vaccine with the trade nameDengvaxia available, it may not provide 100% protection to vaccinees. In addition, the presence of insecticide resistance in Ae. aegypti populations has been reported in several provinces of Thailand. These reasons have led to ineffective prevention and control of the diseases. Therefore, the present study aimed to determine the efficacy of essential oils from seven Thai medicinal plants against the insecticide-resistant Ae. aegypti mosquito from three dengue-risk northern provinces in Thailand. Three northern provinces: Chiang Mai (Hang Dong and San Sai), Tak (Mueang and Ban Tak) and Pitsanulok (Mueang and Nakhon Thai) were chosen for the collection of Ae. aegypti larvae in 2015 based on dengue case report. The mosquito larvae were reared to adults in laboratory. F1 progeny of unfed female aged 3-5 days was tested for resistance status to five pyrethroid insecticides used currently for mosquito control: 0.15% Cyfluthrin, 0.22% Cypermethrin, 0.05% Deltamethrin, 0.05% Lambda-cyhalothrin and 0.75% Permethrin by the WHO susceptibility test. Seven Thai medicinal plants: Citrus hystrix (kaffir lime), Cymbopogon citratus (lemongrass), Murraya siamensis (prong fah), Ocimum americanum (hairy basil), Ocimum basilicum (sweet basil), Ocimum sanctum (holy basil), Piper betel (betel) were extracted by hydrodistillation for essential oils. Each essential oil was then prepared in concentrations of 0.25%, 0.5%, 1.0%, 2.0%, 4.0% and 8.0% in acetone. Those were tested against the insecticide-resistant Ae. aegypti mosquito by modification of the WHO susceptibility test. The two most effective essential oils were developed to an aerosol spray for testing against the insecticide-resistant mosquitoes (100 females with 50 fed and 50 unfed) that were released into the test room (33 m3). The mortality rate of mosquitoes was observed at 24 hours. The insecticide susceptibility test resulted in the mortality rates of mosquitoes from Chiang Mai, Tak and Pitsanulok of less than 80% at 24 hours (<80% = resistance). The essential oils from C. citratus, O. americanum, M. siamensis, O.basilicum, C. hystrix, P. betel and O. Sanctum provided insecticidal activity against the mosquitoesfrom all three provinces with the significant mortality rates of 100%, 100%, 100%, 100%, 100%, 98-100% and 89-100% at minimal concentration of 0.5%, 0.5%, 2.0%, 4.0%, 8.0%, 8.0% and 8.0% at 24 hours, respectively (P<0.05). Aerosol spray containing 5.0% concentration of C. citratus or O. americanum provided a 100% mortality rate of the insecticide-resistant mosquitoes from the three provinces at 24 hours. This study demonstrated that the essential oils from seven Thai medicinal plants were effective against the insecticide-resistant Ae. aegypti mosquito from all three dengue-risk provinces. C. citratus and O. americanum are potentially promising plant sources for alternative compounds to pyrethroids for the control of the vector of dengue fever and zika fever. The knowledge obtained from this study was transferred to public and will be used to develop further as more effective anti-mosquito products for being a part of the sustainable prevention and control program of the Aedes mosquito-borne diseases.
