บทคัดย่อ (EN): It is well accepted that somatic embryogenesis is an ideal system for in vitro plant regeneration. During somatic embryogenesis, some morphological and biochemical changes occur in response to alterations in gene expression patterns. One of the best identified genes associated with somatic embryogenesis induction is Somatic Embryogenesis Receptor Kinase (SERK) gene. SERK gene belongs to the plant receptor kinase superfamily which encodes a leucine-rich repeat receptor-like kinase. Heveabrasiliensis is one of the recalcitrant species for in vitro regeneration, achieved mostly only through somatic embryogenesis. Although, notable success has been achieved by transgenic approaches via somatic embryogenesis, effective utilization of the system need better understanding of the process. Therefore, the present study is aimed to clone and characterize a SERK gene present in Hevea (clone RRII 105) and analyse its expression during somatic embryogenesis. The genomic and cDNA sequences encoding SERK gene in Hevea was amplified with gene specific primers designed based on the consensus sequences reported in other crops. Under optimum PCR conditions, a single band of approx. 0.6 kb size was amplified from genomic DNA, while cDNA yielded a 0.4 kb fragment and both amplicons were cloned and sequenced. The partial HbSERK genomic sequence obtained was 612 bp with one intron and cDNA sequence was 408 bp in length end encoding 135 amino acids. Homology search and sequence analysis demonstrated a high degree of identity with SERK genes from other plant species. Functional validation of cloned HbSERK in somatic embryogenesis was performed by comparing the sequence with AtSERK1 using Clustal W program and results showed 78% similarity in nucleotide sequence while, at the protein level, 99% identity with AtSERK1 sequences. Results of expression analysis revealed higher expression of HbSERK gene in embryogenic callus. In contrast, no expression was detected in mature leaf indicating that HbSERK is associated with induction of somatic embryogenesis. To our knowledge, this is the first report of the characterization of a SERK gene from H. brasiliensis and the sequence was deposited to the GenBank under the accession number KJ451561.
