บทคัดย่อ (EN): This research study investigates the biological production of ethanol from glycerol as a sole substrate with mixed cultures or pure cultures used as microbial seed. Candida tropicalis and Zymomonas mobilis were not able to convert glycerol to ethanol but the mixed cultures from a full scale anaerobic sludge blanket granule of wastewater treatment process did. The simple medium containing mineral salts and glycerol as a sole carbon source was more suitable for the ethanol production than the supplement medium containing both yeast extract and tryptone. The microbial growth, glycerol utilization, and ethanol production exhibited reverse trend with the glycerol concentration at its concentration greater than 45 g/L. Optimal conditions where the maximal ethanol yield (0.95 mol ethanol/mol glycerol) in the simple medium are observed the initial glycerol concentration of 45g/L, incubation temperature at 30 C, pH 7, mixed culture as the seed, and after 56 hr fermentation, 16S rDNA sequence analysis of microbial samples at the optimal condition after 72 hr fermentation with the highest ethanol concentration indicated that 95 percents of the 19 representative clones based on RFLP patterns were in the class Gamma-proteobacteria and the less was in the class Beta-proteobacteria. The dominant microbes in the class Gamma-proteobacteria were closely related to Enterobacter (47.5%) and Klebsiella (47.5 %), while those in the class Gamma-proteobacteria were closely related to genus Thauer.
