บทคัดย่อ (EN): The role of neutral/alkaline invertases (NA-Invs) in determining the sink strength of laticifers and final rubber productivity has been well described in Hevea brasiliensis (Hevea hereafter). However, nothing was known about the relevant gene(s) encoding such NA-Invs in Hevea. In this work, we cloned and functionally characterized the cDNA sequences that code for three NA-Invs expressed in the latex. The three cDNAs, designated HbNIN1, HbNIN2 and HbNIN3, predicted three homologous proteins sharing 12 highly conserved structural domains typical of plant NA-Invs. HbNIN1 and HbNIN2 shared 95.2% amino acids identity and had similar genomic structures containing 4 exons and 3 introns, whereas HbNIN3 was clustered into a different clade, having 7 exons and 3 introns. The HbNIN2 recombinant protein expressed in E. coli showed optimal enzymatic activity at slightly alkalic pH range. By real-time RT-PCR, the expression characters of the three genes revealed to be quite different in planta. In virgin Hevea trees, the expressions of HbNIN1 and HbNIN2 were markedly regulated by tapping, but the patterns were distinct, with HbNIN1 being down-regulated and HbNIN2 being up-regulated. By comparison, the expression of HbNIN3 was less effected by tapping. Wounding alone up-regulated the expressions of HbNIN2 and HbNIN3, but repressed that of HbNIN1. Among the seven hormones examined, the expressions of all three genes were induced by JA and 2,4-D, but were repressed by ABA. In addition, HbNIN3 expression was markedly up-regulated by Ethrel (an ethylene generator). The expressional patterns of these HbNIN genes in response to the treatments of hormones and wounding were further supported by the existence of relevant cis-elements in their promoters. The 454 EST-based expression analyses demonstrated that all three HbNIN genes displayed a predominance expression in the latex, and the expression of HbNIN2 was much more abundant than the other two genes. In addition, N/A Invs were purified from the latex to discernable band(s) revealed by SDS-PAGE and Coomassie blue staining, and MALDI-TOF/TOF tandem mass spectrometry identified that the partially purified NA-Inv was encoded by HbNINI2. In addition, the biochemical characters of the partially purified latex invertase strengthened its traits as NA-Inv, and in situ hybridization revealed the presence of HbNIN2 transcripts in the laticifers. In brief, this work not only reports the cloning and expressional characters of the first two Inv genes in Hevea, but identifies the major member (HbNIN2) in the latex, thus providing a breakthrough point for studying the regulation of sucrose catabolism in the latex and rubber productivity.
