บทคัดย่อ (EN): Rubber is propagated by grafting buds of selected clones on to unselected seeding rootstocks raised in polybags using young budding technique. The growth of rubber seedlings in marginal dry areas of Sri Lanka is slow and therefore they generally take 5-6 months to reach to the buddable stage. Moreover, shoot die-back condition has been observed in polybagged young budded rubber especially under hot and dry weather conditions, partly due to failure of the root system at high temperature. Therefore, it is mandatory to find ways to improve growth and development of rubber seedlings while modifying the root architecture suitable to perform well under heat or drought stress conditions prevailing in the marginal and dry areas of Sri Lanka. Nitric oxide (NO) is a bioactive molecule that functions as a signaling molecule as well as a phytohormone in numerous physiological processes in plants from seed germination up to senescence. It also plays an important role in both biotic and abiotic stress tolerance in plants. Sodium nitroprusside (SNP) has been widely used as an exogenous NO donor to improve growth and development of plants, especially under stress conditions. In the present study, the effect of NO donor SNP on the root architecture and growth characteristics of polybagged rubber seedlings were investigated. Rubber seedlings, four and twelve weeks after transplanting into polybags raised under nursery conditions, were used for the study. Twenty days old rubber seedling were soaked in 200 µmol and 500 µmol solution of SNP for 6hrs and 24hrs respectively prior to transplanting into polybags. Mock treated seedlings were soaked in water. Pretreatment of rubber seedling with SNP greatly improved the root architecture by increasing the number and the length of lateral roots while enhancing other growth parameters viz., stem diameter, shoot height, leaf area and chlorophyll content in a dose-dependent manner. Pretreatment of rubber seedlings in 500 µm SNP for 6 hours was found to be most effective for overall growth performance. The improved growth characteristics of rubber seedlings could be directly attributed to the beneficial effect of SNP as a NO donor in modifying phytohormone signaling pathways, enhancing antioxidant enzyme activity and thereby increase in nutrient uptake and effective allocation of photoassimilates.
