บทคัดย่อ: Bacteria in the genus Vibrio are ubiquitous in marine and estuarine ecosystems. Diversity of these bacteria in tropical areas are not clearly understood. The objectives of this study were to investigate diversity of total Vibrio spp. including V. parahaemolyticus and plankton in aquatic and coastal areas of Thailand. Water samples and plankton were collected monthly from aquatic and coastal areas of Thailand. At Andaman Tarutao Island, water samples and plankton were collected at Son Bay and Reusri Bay from January to April and November to December 2008. At Yor Island (Songkhla lake), water samples and plankton were collected from 2 stations (station A and B) from January to December 2008. Water temperature, salinity and BOD were also determined. Enumeration of bacteria was performed by direct-plating method and filtration method and Vibrio spp. and V. parahaemolyticus were cultivated on TCBS agar and CHROMagar Vibrio respectively. At Tarutao Island, the largest total number of total Vibrio spp. and V. parahaemolyticus in both areas were detected in April when the water temperature was highest. Salinity did not change significantly but the combination of salinity and a high temperature at that time might affect the plankton. In both areas, the total number of plankton dramatically dropped in April and this was correlated to an increase of BOD. Therefore, the abundance of the Vibrio population detected in both areas might be due to the increase in organic compounds released by the dying plankton that supported the growth of Vibrio spp. Statistic analysis at Son Bay revealed high correlation between Vibrio spp. and BOD (r=0.917, p=0.41). At Reusri Bay, relation between V. parahaemolyticus and temperature was in medium correlation (r=0.733, p=0.049). There was no obvious relationship between the kind of either phytoplankton or zooplankton and number of Vibrio spp. observed in both areas. Diversity of Vibrio spp. was determined by DGGE technique. The diversity in Vibrio community at Son Bay and Reusri Bay was different. At Yor Island the total number of Vibrio spp. and V. parahaemolyticus in station A and B were highest in July and June, respectively. Salinity in both stations was between 0-35 ppt. Statistic analysis at station A indicated high correlation between number of Vibrio spp. and V. parahaemolyticus and salinity (r=0.822, p=0.01 และ r=0.879, p =0.01 respectively). At station B relation between number of Vibrio spp. and V. parahaemolyticus and salinity were in medium correlation (r=0.716, p =0.04 และ r=0.773, p =0.002 respectively) which was the same as correlation between number of Vibrio spp. and V. parahaemolyticus and temperature (r=0.662, p =0.018 และ r=0.690, p =0.014 respectively). Interestingly, in November which salinity was 0 ppt., V. parahaemolyticus and V. cholerae were isolated from station A and station B respectively at Yor Island. Since Yor Island is close to community, this indicates that water in this area must be contaminated with pathogenic Vibrio spp. In addition, it is indicated that some strains of V. parahaemolyticus could adapt themselves to survive in environment that lack of sodium chloride. There was no obvious relationship between the kind of either phytoplankton or zooplankton and number of Vibrio spp. observed at Yor Island. Diversity of Vibrio spp. at station A and station B was similar.
