บทคัดย่อ: ผลหม่อนเป็นผลไม้ที่มีฤทธิ์บำรุงสายตาในการแพทย์พื้นเมือง แต่ยังไม่พบข้อมูลทางวิทยาศาสตร์ที่จะ ยืนยันสรรพคุณดังกล่าวในปัจจุบัน ดังนั้นการศึกษาครั้งนี้จึงมุ่งศึกษาฤทธิ์ป้องกันต้อกระจกในผู้สูงอายุ (senile cataract) และต้อกระจกจากเบาหวาน (diabetic cataract) ของผลหม่อน ตลอดจนพัฒนาผลหม่อนเป็น อาหารสุขภาพบำรุงสายตาป้องกันต้อกระจก ในการศึกษาฤทธิ์ป้องกันต้อกระจกของผลหม่อนอบแห้ง และสาร สกัดแอลกอฮอล์ของผลหม่อนอบแห้งในแบบจำลองที่เหนี่ยวนำให้เกิดต้อกระจกในผู้สูงอายุด้วย selenite พบว่า ทั้งผลหม่อนอบแห้งและสารสกัดแอลกอฮอล์ของผลหม่อนอบแห้งสามารถป้องกันการเกิดต้อกระจกใน ผู้สูงอายุได้ กลไกการออกฤทธิ์น่าจะเกี่ยวข้องกับการลดความเครียดออกซิเดชัน ข้อมูลจากการศึกษาฤทธิ์ ป้องกันต้อกระจกจากเบาหวานในแบบจำลองที่เหนี่ยวนำด้วย streptozotocin แสดงให้เห็นว่าทั้งผลหม่อน อบแห้งและสารสกัดแอลกอฮอล์ของผลหม่อนอบแห้งสามารถป้องกันต้อกระจกจากเบาหวานได้ โดยออกฤทธิ์ ผ่านการลดความเครียดออกซิเดชัน การยับยั้งการทำงานของเอนไซม์สโดสรีดักเตส (Aldose reductase) และการศึกษาการยับยั้งการอักเสบ พบว่าประสิทธิภาพในการป้องกันต้อกระจกของผลหม่อนอบแห้ง และสาร สกัดแอลกอฮอล์ของผลหม่อนอบแห้งไม่ต่างกัน จึงได้ทำการพัฒนาผลิตภัณฑ์สุขภาพบำรุงสายตาต้นแบบ โดย ใช้ผลหม่อนอบแห้งเป็นวัตถุดิบ น้ำสกัดจากผลหม่อนอบแห้งชนิดเข้มข้นที่พัฒนาขึ้นจะมีสาร total phenolic compounds อยู่ในช่วง 715.83-865.83 mg of gallic acid equivalent (CGE) ต่อ 100 กรัมของตัวอย่าง ปริมาณสารแอนโทไซยานิน 589.50-738.47 CGE ต่อ100 กรัมของตัวอย่าง ปริมาณเควอซิติน 9.39-11.07 มิลลิกรัมต่อ 100 กรัมของตัวอย่าง มีปริมาณรูติน 6.90-7.53 มิลลิกรัมต่อ 100 กรัมของตัวอย่าง ข้อมูลจาก การทดสอบฤทธิ์ทางเภสัชวิทยาในสัตว์ทดลองที่จำลองภาวะต้อกระจกในผู้สูงอายุ พบว่า สัตว์ทดลองที่ได้รับน้ำ สกัดจากผลหม่อนอบแห้งชนิดข้มข้นขนาด 2 และ 10 มิลลิกรัมต่อกิโลกรัมน้ำหนักตัว เป็นเวลา 14 วัน จะลด ภาวะต้อกระจกได้มากกว่ากลุ่มที่ไม่ได้รับน้ำสกัดจากผลหม่อน นอกจากนั้นยังพบว่าสัตว์ทดลองที่ได้รับการ เหนี่ยวนำให้เป็นต้อกระจกจากเบาหวานด้วย streptozotocin และได้รับน้ำสกัดจากผลหม่อนอบแห้งชนิด เข้มข้นขนาด 10 มิลลิกรัมต่อกิโลกรัมน้ำหนักตัว จะลดการเกิดต้อกระจกจากเบาหวาน อย่างไรก็ตามควรมี การศึกษากลไกการออกฤทธิ์ที่แน่นอนของน้ำสกัดจากผลหม่อนอบแห้งชนิดเข้มข้น ก่อนที่จะนำไปศึกษาใน clinical trial เพื่อนำไปใช้ต่อไป
บทคัดย่อ (EN): Mulberry is the fruit which is reputed for eye preservation in traditional folklore but no supported scientific evidence is available until now. Therefore, this study aimed to determine the protective effect against senile and diabetic cataracts of mulberry fruit and to develop mulberry fruit as the functional food for preventing cataract. In the study which aimed to determine the preventive effect against senile cataract induced by selenite of dried mulberry fruit powder and alcoholic extract of dried mulberry fruit powder, it was found that both dried mulberry fruit powder and alcoholic extract of dried mulberry fruit powder showed the anti-cataract effect. The mechanism of action might be associated with the decreased oxidative stress. Data obtained from the study about the preventive effect against diabetic cataract of dried mulberry fruit powder and alcoholic extract of dried mulberry fruit powder in animal model induced by streptozotocin also demonstrated the antidiabetic cataract of both dried mulberry fruit powder and alcoholic extract of dried mulberry fruit powder. The mechanism of action might involve the reduction of oxidative stress and the suppression of aldose reductase and inflammation. The protective efficiency of dried mulberry fruit and alcoholic extract of dried mulberry fruit powder didn’t show much difference so we developed the prototype product of anticataract functional food by using dried mulberry fruit powder as resource. Concentrated mulberry fruit extract which had been developed contained total phenolic compounds, anthocyanin, quercetin and rutin at the following concentrations; 715.83 - 865.83 mg of gallic acid equivalent/100 g of samples, 589.50 - 738.47 CGE/100 g sample, 9.39 - 11.07 mg/100 g sample and 6.90 - 7.53 mg/100 g sample respectively. Data obtained from the evaluation of pharmacological activity in animal model of senile cataract showed that animals which subjected to the concentrated mulberry fruit extract at doses of 2 and 10 mg/kg for 14 days decreased senile cataract when compared to the non-treated animals. It was also found that the concentrated mulberry fruit extract at dose of 10 mg/kgBW decreased diabetic cataract in animal model induced by streptozotocin. However, further investigation about the precise underlying mechanism should be carried out before moving forward to clinical trial study and application.
