บทคัดย่อ (EN): In the last twenty years, Corynespora leaf fall disease (CLFD) has become one of the most serious leaf diseases, severely affecting the international rubber industry. It is currently responsible for 20-25% of the economic losses in natural rubber production. CLFD has been reported by the rubber producing countries in Asia, which account for 94% of the world’s natural rubber production. Identification of resistant clones is a major strategy in the management of CLFD in rubber. On screening the clones of rubber under natural conditions, we found that ‘IAN873’ was a resistant germplasm, and ‘PR107’ was susceptible to CLFD. In order to illustrate the resistant and susceptible response of the rubber tree (Hevea brasiliensis) infected with Corynespora cassiicala at a transcriptional level, gene expression profiles in leaves of rubber tree, inoculated with fungal spore suspension, were tested using cDNA-AFLP technology. Selective amplifications with 96 primer combinations allowed the visualization of 4,800 transcript-derived fragments (TDFs) in infected leaves, and 560 of them were found to be differentially expressed (306 induced and 254 repressed). 159 TDFs were cloned and sequenced, of which 116 were shown to have known functions and had predicted gene products by searching the GenBank database through BLAST. Based on the conclusion of the cDNA-AFLP analysis, four resistance genes were cloned by RT-PCR and PCR technique. There are a plant resistance NPR gene family member HbNPR1, a haloalcanoic acid dehalogenase super family member HbVSP2 and resistance gene analog members HbRGH2 and HbRGA-1. The expressed patterns of four representative TDFs were further confirmed by real-time PCR analysis. These differentially expressed genes may be involved in the processes of the anti/flu disease and our study confirms the effectiveness of isolation and cloning disease resistance genes.
