บทคัดย่อ (EN): Background: Infection of Dengue, Chikungunya and Zika viruses can cause similar symptoms. Performing laboratory test for each virus is time consuming. Multiplex real-time RT-PCR is a method which has high sensitivity/specificity, low cross-reaction and can detect several pathogens simultaneously in one test. This study was aimed to develop the multiplex real-time RT-PCR for Dengue, Chikungunya and Zika virus detection in Thai clinical samples. Materials/Methods: Multiplex real-time RT-PCR were developed with primers and probes specific to each viruses and test with laboratory strains of Dengue1, Dengue2, Dengue3, Dengue4, Chikungunya and Zika viruses to optimized and determine the limit of detection. Japanese encephalitis, Yellow fever and West Nile virus were tested for cross reaction. A total of Thai 148 known positive samples and 23 known negative samples for Dengue, Chikungunya and Zika virus infection were used to evaluate this test. The positive samples consisted of 80 Dengue (20 for each serotype), 23 Chikungunya and 45 Zika. Results: All laboratory strains were detected with the developed method. Limit of detection for Dengue1, Dengue2, Dengue3, Dengue4 viruses were approximately 0.46, 8, 8 and 17.2 FFU/ml, respectively. Additionally, limit of detection for Zika and Chikungunya viruses were approximately 89 and 21 copies/ml, respectively. No cross-reaction occurred with Japanese encephalitis, Yellow fever and West Nile viruses. The detection sensitivity of clinical Dengue and Chikungunya was 100%, while that for Zika was 95.56%. The specificity was 100% for all three viruses and no false positive result occurred in this study. Discussion and Conclusion: The developed method showed satisfactory results with acceptable sensitivity and specificity. Negative results in some Zika-infected samples could possibly due to very low titer of virus in those specimens.
